PNC-27
PNC-27 is an experimental peptide studied primarily for its potential anti-cancer properties, particularly its ability to selectively kill cancer cells in laboratory settings.
What it is
PNC-27 is a synthetic peptide derived from a fragment of the p53 protein — specifically the region of p53 that binds to HDM-2 (also called MDM-2), a protein that’s overexpressed on the surface of many cancer cell membranes. It’s attached to a membrane-penetrating sequence to help it reach and interact with cell membranes.
How it’s thought to work
- p53 is a well-known tumor-suppressor protein; many cancers either mutate p53 or overexpress HDM-2 to inactivate it
- Unlike normal p53, which works inside the cell, PNC-27 is designed to act at the cell membrane
- It reportedly binds to HDM-2 on the surface of cancer cells specifically (HDM-2 is much less present on normal cell membranes)
- This binding is theorized to create pores in the cancer cell membrane, causing rapid necrosis (cell death through membrane rupture) rather than apoptosis
Research status
- Studied mainly in in vitro (petri dish/cell culture) settings and some animal models by a small number of research groups (notably work published by Pazo Biotech researchers)
- Early studies suggested selectivity for cancer cells over normal cells, since normal cells have much lower HDM-2 surface expression
- No completed, published human clinical trials establishing safety or efficacy exist in the mainstream literature
- Not FDA-approved, not a recognized cancer treatment
Current status and caveats
- Like SS-31 and Adipotide, PNC-27 is sold in the unregulated “research peptide” marketplace, typically labeled “for research use only, not for human consumption”
- No quality control, dosing, or purity standards apply to these products
- There is no legitimate medical pathway to obtain or use PNC-27 as a cancer treatment currently — self-administering an unapproved, unregulated peptide for cancer (a serious disease needing real oversight) carries significant risk and no verified benefit
